ĐÁNH GIÁ ĐA DẠNG DI TRUYỀN TẬP ĐOÀN HOA LAN HOÀNG THẢO PHI ĐIỆP (DENDROBIUM ANOSMUM) BẰNG GIẢI TRÌNH TỰ VÙNG ITS (INTERNAL TRANSCRIBED SPACER)

Abstract

The Dendrobium anosmum orchids available on the market are very diverse and complex and it is difficult to distinguish between one variety and another because their appearance is very similar before flowering. In this study, 118 samples of Dendrobium anosmum from were collected from different regions were amplified and successfully resolved for ITS1, 5.8S and ITS2 (Internal Transcribed Spacer) regions with lengths from 700 to 800 bp. Based on the attained results of the sequencing, ITS has identified two distinct varieties, L28 (Dendrobium aphyllum) and L35 (Dendrobium parishii) varieties and other varieties belong to the Dendrobium anosmum species. Comparing the ITS segments, most Vietnamese native Dendroium anosmum have high similarity to compare with the world's samples. Some different nucleotide positions ranged from the 592-646 segments, so it is difficult to distinguish the differences between the cultivars within the same species. Therefore, in order to accurately identify each variety in the same species of Dendrobium anosmum in Vietnam, it is necessary to continue to use other advanced techniques such as SSR or ISSR marker or whole-genome sequencing of varieties in the group.

Các giống hoa lan Hoàng Thảo Phi Điệp (Dendrobium anosmum) trên thị trường rất đa dạng và phức tạp, rất khó để phân biệt giữa giống này với giống khác vì đặc điểm hình thái bên ngoài của chúng rất giống nhau trước khi phát triển hoa. Trong nghiên cứu này, 118 mẫu giống Hoàng Thảo Phi Điệp (Dendrobium anosmum) thu thập ở các vùng khác nhau đã được khuếch đại và giải thành công vùng ITS1, 5.8S và ITS2 (Internal Transcribed Spacer) với chiều dài từ 700 - 800 bp. Dựa trên kết quả giải trình tự vùng ITS đã xác định được 02 mẫu giống khác biệt là mẫu giống L28 (Hạc vĩ - Dendrobium aphyllum) và mẫu giống L35 (Hoàng Thảo trầm - Dendrobium parishii), các mẫu giống còn lại đều thuộc loài Dendrobium anosmum. Qua kết  quả so sánh trình tự đoạn ITS của các mẫu giống hoa lan Hoàng Thảo Phi Điệp bản địa Việt Nam và thế giới cho thấy chúng có độ tương đồng cao so với các mẫu của thế giới, chỉ có sự sai khác ở một số vị trí nucleotide từ đoạn 592-646 bp và được phân thành các nhóm khác nhau. Dựa vào trình tự sai khác chỉ có thể nhận dạng được một số mẫu giống trong loài. Vì vậy, muốn xác định chính xác từng giống trong cùng một loài Dendrobium anosmum của Việt Nam, cần tiếp tục sử dụng thêm các chỉ thị khác như SSR hoặc ISSR hoặc giải trình tự hệ gen của các giống trong nhóm.
