TINH SẠCH IMMUNOGLOBULIN G (IgG) TỪ MÁU HEO (Sus domesticus)

Abstract

The study of Immunoglobulin G (IgG) purification from pig blood (Sus domesticus) was performed on three protein precipitation solvents, ethanol, acetone and ammonium sulfate. Then the best solvent and precipitation rate will be selected for gel filtration chromatography on Sephadex G-100 gel and ion exchange chromatography on DEAE CL-6B gel. The analytical results showed that when precipitated with ehtanol, acetone and amonium sulfate at different concentrations, the ammonium sulfate precipitate with a concentration of 30% - 40% was optimal and was chosen to perform gel filtration chromatography on Sephadex gel. G-100. The results obtained a single peak of F (9-13) with a protein content of 1,261 mg with a recovery efficiency of 27% and fluorescence results showed that the obtained IgG content was 216 mg. Fraction F (9-13) of gel filtration chromatography continued to be negative ion exchange chromatography on DEAE Sepharose CL-6B gel, obtained two peaks F1 and F2 with F1 eliminated most albumin, F1 had protein is 985 mg with recovery efficiency of 6.11% and obtained IgG content is 120 mg.

Nghiên cứu tinh sạch Immunoglobulin G (IgG) từ máu heo (Sus domesticus) được thực hiện trên ba dung môi kết tủa là ethanol, acetone và ammonium sulfate. Sau đó dung môi và tỷ lệ tủa tốt nhất sẽ được chọn để tiến hành sắc ký lọc gel trên gel Sephadex G-100 và sắc ký trao đổi ion trên gel DEAE CL-6B. Kết quả phân tích cho thấy khi tủa với ehtanol, acetone và amonium sulfate ở các nồng độ khác nhau thì kết tủa ammonium sulfate với nồng độ 30% - 40% là tối ưu và được chọn để tiến hành sắc ký lọc gel trên gel Sephadex G-100. Kết quả thu được một đỉnh duy nhất là F (9 - 13) có hàm lượng protein là 1.261 mg với hiệu suất thu hồi đạt 27% và kết quả nhuộm huỳnh quang cho thấy hàm lượng IgG thu được là 216 mg. Phân đoạn F (9 - 13) của sắc ký lọc gel tiếp tục được sắc ký trao đổi ion âm trên gel DEAE Sepharose CL-6B, thu được hai đỉnh F1 và F2 với F1 loại bỏ được phần lớn albumin, F1 có hàm lượng protein là 985 mg với hiệu suất thu hồi đạt 6,11% và hàm lượng IgG thu được là 120 mg.
