NGHIÊN CỨU KHẢ NĂNG NHÂN GIỐNG LAN HÀI VIỆT NAM (Paphiopedilum vietnamese) BẰNG KỸ THUẬT IN VITRO

Abstract

Paphiopedilum vietnamese is one of the endemic species in Vietnam. Selection of fruit after 9 months pollination. Seed germinated on germination MS medium supplemented with 30 g/l sugar, 1 g/l activated charcoal, 5.5 g/l agar, 150 ml/l coconut water. Ability of propagation of Paphiopedilum vietnamese from seed embryos showed that regeneration and shoot multiplication on the medium included BA 4 mg/l or BA 4 mg/l combination with Kinetin 0.6 mg/1 in MS medium have been sugar 30 g/l, 1 g/l activated carbon, 5.5 g/l agar, 150 ml/l of coconut water, obtained 86.2, average height reached 2.71 cm, good quality. The best growth and rooted plant medium as is MS medium supplemented with 30 g/l sugar, 5.5 g/l agar, 150 ml/l coconut water and 1 mg/l NAA or 1mg/l IAA.

Lan hài Việt Nam (Paphiopedilum vietnamese) là một trong những loài lan đặc hữu của Việt Nam. Quả thụ phấn sau 9 tháng được lựa chọn để tiến hành thí nghiệm. Phôi hạt nuôi nảy mầm trên môi trường MS bổ sung thêm 30 g/l đường, 1 g/l than hoạt tính, 5,5 g/l agar, 150 ml/l nước dừa. Nhân nhanh loài lan hài Việt Nam từ phôi hạt cho thấy khả năng tạo đa chồi tốt khi bổ sung chất điều tiết sinh trưởng BAP 4 mg/l hoặc BAP 4 mg/l kết hợp Kinetin 0,6 mg/l vào nền môi trường MS có đường 30 g/l, than hoạt tính 1 g/l, agar 5,5 g/l, nước dừa 150 ml/l, thu được 86,2 chồi, chiều cao chồi trung bình đạt 2,71 cm, chất lượng chồi tốt. Rễ lan hài Việt Nam phát sinh tốt nhất trong điều kiện môi trường MS bổ sung đường 30 g/l, agar 5,5 g/l, nước dừa 150 ml/l và NAA hoặc IAA nồng độ 1 mg/l.
