ỨNG DỤNG KỸ THUẬT MULTIPLEX RT-PCR PHÁT HIỆN Cymbidium mosaic virus (CymMV) VÀ Odontoglossum ringspot virus (ORSV) GÂY HẠI TRÊN CHI ĐĂNG LAN (Dendrobium)

Abstract

Dendrobium orchids are beautiful and precious cut flowers with high economic potential. However, virus diseases in orchids is a major barrier causing very negative impact on the orchid growing industry. The purpose of this study was to apply multiplex RT-PCR to detect and survey the infection rate of CymMV and ORSV in the orchid genus Dendrobium. The study results demonstrated that the virus detection capacity from the multiplex RT-PCR completely identical to the virus detection results from the simple RT-PCR. Thus, the results showed that the multiplex RT-PCR applied in this study was a rapid, simple, accurate and reliable technique. In addition, the results of using multiplex RT-PCR on 40 Dendrobium orchid samples collected from the orchid garden detected 26 CymMV infections, 2 ORSV infections, two of which 2 ORSV infections. Two of which were co-infected with CymMV and ORSV. The archived results of this study were useful information contributing to virus research on the genus Dendrobium orchid in Vietnam.

Lan là một trong những loài hoa đẹp, quý và có tiềm năng kinh tế rất cao. Tuy nhiên, bệnh do virus gây ra trên lan là rào cản lớn đã tác động rất tiêu cực đến kinh tế ngành trồng lan. Mục tiêu của nghiên cứu này là áp dụng quy trình multiplex RT-PCR để phát hiện và khảo sát tỉ lệ nhiễm CymMV và ORSV trên các mẫu lan Dendrobium. Kết quả nghiên cứu đã chứng minh khả năng phát hiện virus từ quy trình multiplex RT-PCR hoàn toàn trùng khớp với kết quả phát hiện virus từ quy trình simplex RT-PCR. Như vậy, kết quả đã chỉ ra rằng, multiplex RT-PCR được ứng dụng trong nghiên cứu này là kỹ thuật nhanh chóng, đơn giản, chính xác và đáng tin cậy. Thêm nữa, kết quả sử dụng quy trình multiplex RT-PCR trên 40 mẫu lan Dendrobium thu thập từ vườn thực địa đã phát hiện 26 mẫu nhiễm CymMV, 2 mẫu nhiễm ORSV, trong đó có 2 mẫu nhiễm ORSV cũng là mẫu nhiễm đồng thời CymMV và ORSV. Kết quả đạt được của nghiên cứu này là những thông tin hữu ích góp phần cung cấp thêm thông tin về nghiên cứu virus gây hại trên chi đăng lan Dendrobium.
