Abstract

Potato (Solanum tuberosum L.) is a globally significant crop, including in Vietnam. Traditional breeding methods for potatoes are often challenging due to the complexity of the potato genome. While precision breeding techniques like gene transformation and editing offer advantages, they heavily rely on efficient in vitro culture and shoot regeneration processes. The Marabel potato variety, popular in production for its outstanding characteristics, has been understudied in terms of in vitro culture. This study investigated key factors influencing callus formation in Marabel potatoes and determined an optimal culture medium containing 2,4-D (1.0 mg/L) and Zeatin (1.0 mg/L), achieving 90% efficiency. Shoot regeneration from callus reached 31.7% using a medium combining Zeatin (1.0 mg/L) and NAA (0.1 mg/L). These findings provide a crucial foundation for future gene transfer and editing research in the Marabel potato variety.
